    • 10 kb biotin-ended DNA is prepared using long expand PCR system (Roche Diagnostics) on lambda DNA with designed primers: 5′-biotin- CTgATgAgTTCgTgTCCgTACAACTggCgTAATC-3′ and 5′-(biotin)- ATACgCTgTATTCAgCAACACCgTCAggAACACg-3′. 2.4 kb is prepared by PCR from Bluescript plasmid with designed primers: 5′-biotin-AgTgCTgCCATAACCATg Ag-3′ and 5′-(biotin)-CgAgATAgggTTgAgTgTTg-3′.
