Unexpectedly, the structure revealed that the GRAM domain has a PH domain fold. Several of the structural determinants of PI substrate specificity were accurately inferred based on the structure of the PTP domain. All missense disease mutations in MTM1 and MTMR2 reported at the time were mapped to the structure.
