The most sensitive region of a beam quadrant with respect to refractive index changes is very close to the focus of the microscope objectives. This is the region where the beam quadrants just can be optically separated, considering the diffraction limit of about 200 nm. The region is determined by a complicated three-dimensional optical transfer function defined by the numerical apertures of the objectives and the position of the detection fiber. In total, eight such regions are present for each of the four beam quadrants, one in front of and one after the focus.
