This study found that prolonged labor was associated with atopy, and severe early respiratory infection and induced labor with bronchial hyperresponsiveness after trying to sift through a myriad of perinatal events, some quite transient, which may have had an impact on allergic risk. The risk factors were retrospectively analyzed, while allergic outcomes were measured by methacholine challenge and skin prick testing in addition to subjective surveys. Over 500 patients were studied at the 20-year follow-up of a birth cohort from the Netherlands.

In this prospective observational cohort, each febrile episode prior to the age of 1 was associated with reduced odds of allergic sensitization as defined by either skin-prick positivity or seroatopy by age 6 or 7. Febrile URIs, in particular, reduced the risk even more, providing evidence that certain early infections may indeed protect from atopy, supporting at least a part of the claim made in the hygeine hypothesis.

Polymorphisms at the CD14 locus were used to segregate 1-year-old infants whose environmental histories were taken and allergic outcomes measured by skin prick, PBMC cytokine production and clinical histories. Amongst other findings, infants with several particular polymorphisms at CD14 were protected from atopic dermatitis if they had a dog in the house, whereas those with another genotype did not.
