Cytochrome c folding pathway: Kinetic native-state hydrogen exchange

Prior measurements of H/D exchange under near native conditions have provided free-energy changes for cooperative unfolding of five structural units in cyt c. Submillisecond time resolution permits characterization of two compact intermediates that form before the native structure. Interrupted refolding measurements provide direct evidence for parallel folding pathways in this bacterial cyt c with a typical class I fold. The obvious question is whether parallel folding pathways also are active in the mitochondrial protein. If not, then it is reasonable to question whether the topology of the native protein is an important determinant of the folding mechanism.
