The relative roles of advanced glycation, oxidation and aldose reductase inhibition in the development of experimental diabetic nephropathy in the Sprague-Dawley rat
-
Soulis-Liparota T, Cooper ME, Dunlop M, Jerums G: The relative roles of advanced glycation, oxidation and aldose reductase inhibition in the development of experimental diabetic nephropathy in the Sprague-Dawley rat. Diabetologia 1995, 38:387-394. This is an original paper with a thoughtful discussion of various mechanisms for development of diabetic nephropathy, concluding that oxidative stress is not involved. The studies were conducted in streptozotocin-diabetic rats receiving low-level insulin therapy for 8 months, an animal model that may be more relevant to chronic human diabetes than the acute, untreated diabetic rat model.

Vitamin E prevents diabetes-induced abnormal retinal blood flow via the diacylglycerol-protein kinase C pathway

Kunisaki M, Bursell SE, Clermont, AC, Ishii H, Ballas LM, Jirousek MR, Umeda F, Nawata H, King GL: Vitamin E prevents diabetes-induced abnormal retinal blood flow via the diacylglycerol-protein kinase C pathway. Am J Physiol 1995, 269:E239-E246. This is an excellent study showing that vitamin E therapy inhibits development of retinopathy in diabetic rats and illustrating that alterations in the protein kinase C pathway in diabetes and in retinal endothelial cells grown in hyperglycemic media are also reversible by vitamin E.

Nitric oxide as a potential bridge between the metabolic and vascular hypotheses of diabetic neuropathy
-
Stevens MJ: Nitric oxide as a potential bridge between the metabolic and vascular hypotheses of diabetic neuropathy. Diabet Med 1995, 12:292-295. This is a thoughtful review developing the relationship between biochemical abnormalities in polyol pathway and protein kinase C activity during hyperglycemia and subsequent alterations in nitric oxide metabolism and vascular tone.

-
Cameron NE, Cotter MA, Hohman TC: Interactions between essential fatty acid, prostanoid, polyol pathway and nitric oxide mechanisms in the neurovascular deficit of diabetic rats. Diabetologia 1996, 39:172-182. This is the latest of a series of papers on alternative approaches to the treatment of neuropathy in acutely and severely diabetic rats. Endothelial relaxation is disturbed by oxidative stress in the diabetic rat heart: Influence of tocopherol as antioxidant
-
Rösen P, Ballhausen T, Bloch W, Addicks K: Endothelial relaxation is disturbed by oxidative stress in the diabetic rat heart: influence of tocopherol as antioxidant. Diabetologia 1995, 38:1157-1168. This study shows that administration of vitamin E protects the rat heart against functional and morphologic damage in diabetes, suggesting a role for vitamin E in the treatment of diabetic cardiomyopathy.
