GUVs were prepared as described previously, except lipids were spread on ITO coated glass slides at an elevated temperature (60°C) to produce uniform films. Phospholipids and cholesterol were purchased from Avanti Polar Lipids (Alabaster, AL). Racemic PSM was purchased from Sigma (St. Louis, MO) and is a mixture of d-erythro and l-threo isomers.

We set a threshold to distinguish dark, solid domains from dark, liquid domains by a decrease in lipid mobility. Specifically, we state that two circular, dark, solid domains do not coalesce within minutes [Fig. 1(e)], and motion of bright, liquid domains is not visible by eye on a back-ground of dark, solid phase in real time.
