    • To further evaluate compounds 2 and 3, we studied their antagonistic activity in two pharmacological in vivo models (for experimental details, see Supporting Information). The first model investigated the antihypertensive response in conscious spontaneously hypertensive rats (SHR), using DuP 753 (losartan) as a reference compound. Compound 2 displayed weak antihypertensive effects compared to DuP 753. Compound 3, however, showed a pronounced hypotensive effect even at very low concentrations (0.001 μmol/kg). Higher concentrations than 0.1 μmol/kg compound 3 could not be given due to adverse cardiac effects. These effects did not seem to be related to parasympatic activity as similar results were obtained in rats pretreated with atropine. In the second model, SHR were constantly infused with Ang I. No dose dependent hypotensive effect was seen for compound 2 or 3, even at a dose of 10 μmol/ kg compound 2 and 1 μmol/kg compound 3. DuP 753 investigated in the latter in vivo model showed a dose dependent hypotensive effect on the mean arterial blood pressure. The antagonistic/ hypotensive effect that compound 3 displays in one of the in vivo models could be argued as being an effect on the renin angiotensin system as compound 3 did not show any dose dependent hypotensive effect during Ang I infusion. This would rather support the agonistic profile seen in the rabbit aorta strips model.
