Chang and coworkers show that the immunodepletion of ε-tubulin blocks centriole duplication in Xenopus egg extracts. Using immuno-electron microscopy, they show that ε-tubulin is localized to the subdistal appendages on the old centriole.

The bld2-1 mutant was isolated in 1975 and fails to assemble full-length basal bodies. A chromosome walk revealed that the BLD2 locus encodes ε-tubulin. ε-tubulin is localized in a new structure around the mature basal bodies.
