Proc Natl Acad Sci USA 1997, 94:7452-7457. The origin of the Friedreich ataxia GAA expansion mutations in the European population is traced down by linkage disequilibrium analysis. The frequency of heterozygote carriers of expansion is directly estimated from the study of 851 individuals.

Campuzano V, Montermini L, Molt MD, Pianese L, Cossé M, Cavalcanti F, Monros E, Rodius F, Duclos F, Monticelli A et al.: Friedreich ataxia: autosomal recessive disease caused by an intronic GAA triplet repeat expansion. Science 1996, 271:1423-1427. The discovery of an intronic GAA expansion as the major cause of Friedreich ataxia was surprising in many regards as the position, sequence and mode of inheritance of the expansion mutation were different from other expansion diseases. The identified gene encoded a novel protein that is conserved through evolution.

Deletion of the yeast homologue of the human gene associated with Friedreich's ataxia elicits iron accumulation in mitochondria
-
Foury F, Cazzalini O: Deletion of the yeast homologue of the human gene associated with Friedreich's ataxia elicits iron accumulation in mitochondria. FEBS Lett 1997, 411:373-377. In these three papers [36••,37•,38;••], the authors demonstrate the involvement of yeast frataxin in iron homeostasis both from the phenotype of the corresponding knock-out and from high copy suppression of another mutant showing slow growth on iron-poor medium. The results suggest that oxidative stress is the primary pathological mechanism in Friedreich ataxia.
