The discovery of an intronic GAA expansion as the major cause of Friedreich ataxia was surprising in many regards as the position, sequence and mode of inheritance of the expansion mutation were different from other expansion diseases. The identified gene encoded a novel protein that is conserved through evolution.

In these three papers [36••,37•,38;••], the authors demonstrate the involvement of yeast frataxin in iron homeostasis both from the phenotype of the corresponding knock-out and from high copy suppression of another mutant showing slow growth on iron-poor medium. The results suggest that oxidative stress is the primary pathological mechanism in Friedreich ataxia.
