Hijacking of host cell IKK signalosomes by the transforming parasite Theileria
-
Heussler VT, Rottenberg S, Schwab R, Kuenzi P, Fernandez PC, McKellar S, Shiels B, Chen ZJ, Orth K, Wallach D et al.: Hijacking of host cell IKK signalosomes by the transforming parasite Theileria. Science 2002, 298:1033-1036. This work describes how constitutive NF-κB activation in Theileria-transformed cells is mediated by the recruitment of the multisubunit IκB kinase, IKK, into large activated foci on the surface of the parasite. IKK signalosome assembly is specific for the transforming schizont stage and is downregulated upon differentiation into the non-transforming merozoite stage.

The Akt/PKB pathway is constitutively activated in Theileria-transformed leucocytes, but does not directly control constitutive NF-κB activation

Heussler VT, Kuenzi P, Fraga F, Schwab RA, Hemmings BA, Dobbelaere DA: The Akt/PKB pathway is constitutively activated in Theileria-transformed leucocytes, but does not directly control constitutive NF-κB activation. Cell Microbiol 2001, 3:537-550. This report demonstrates that the PtdIns-3K-Akt/PKB pathway is constitutively activated in Theileria-transformed cells and contributes to proliferation. A link between the PtdIns-3K-Akt/PKB pathway and constitutive NF-κB activity could be excluded.

Constitutive exclusion of Csk from Hck-positive membrane microdomains permits Src kinase-dependent proliferation of Theileria-transformed B lymphocytes

Baumgartner M, Angelisova P, Setterblad N, Mooney N, Werling D, Horejsi V, Langsley G: Constitutive exclusion of Csk from Hck-positive membrane microdomains permits Src kinase-dependent proliferation of Theileria-transformed B lymphocytes. Blood 2003, 101:1874-1881. This work shows that the uncontrolled proliferation of Theileria-transformed B cells is associated with constitutive activation of the Src family protein kinase Hck, apparently caused by exclusion of its negative regulator, Csk from Hck-positive glycolipid-enriched microdomains.

Characterisation of a cluster of genes encoding Theileria annulata AT hook DNA-binding proteins and evidence for localisation to the host cell nucleus
-
Swan DG, Stern R, McKellar S, Phillips K, Oura CA, Karagenc TI, Stadler L, Shiels BR: Characterisation of a cluster of genes encoding Theileria annulata AT hook DNA-binding proteins and evidence for localisation to the host cell nucleus. J Cell Sci 2001, 114:2747-2754. This paper describes a cluster of Theileria genes, called TashATs, the products of which contain AT-hook DNA-binding motifs. Experiments using antibodies raised against TashATs suggest they may be secreted by the parasite and translocate to the host nucleus.
