Auxin and salicylic acid are shown to modulate the expression of AO in tobacco leaves. These plant hormones also regulate the growth of tobacco plants in a manner consistent with a role for AO in the regulation of growth. Transgenic tobacco plants that express AO in the sense and antisense orientations confirm this view.

Plant Cell 2003, 15:1212-1226. The authors of this paper provide the first molecular signature of ascorbate deficiency in plants. They demonstrate that plant growth and defence may be modulated simultaneously in response to changes in ascorbate availability via changes in the ABA and ABA/GA signalling pathways. The abundance of key transcripts is modulated by ascorbate availability in leaves. Thus, plants sense not only changes in the redox state of their ascorbate pool but also the absolute amount of this major redox buffer.

Using a genetic approach to screen for mutants, the authors identify a new serine-threonine protein kinase, Stt7, in the chloroplast thylakoid membrane. This kinase phosphorylates light-harvesting chlorophyll a/b binding protein (LHC II) and has a plastoquinone-binding site. It links redox signalling, kinase activation and gene expression. Modulation of kinase function by the plastoquinone redox state controls the relative rates of transcription of photosystem I and photosystem II reaction-centre genes. It also regulates systemic signals that convey information on light stress to distant tissues. Stt7 was identified in Chlamydomonas but has clear orthologues in Arabidopsis, opening the door to the elucidation of the nature of chloroplastic redox signalling in higher plants. In particular, this work shows that the kinase structure houses two cysteines that could be reversibly modulated by the redox regulatory protein thioredoxin.
