J Biol Chem 2002, 277:14986-14995. The authors characterise a mutant form of DnaA (R334A) that binds but cannot hydrolyse ATP following initiation of DNA replication. Over-expression of DnaA R334A leads to an over-initiation of replication that is lethal for the cells. This paper demonstrates the importance of regulating the ATP- or ADP-bound forms of DnaA for correct timing of replication initiation.
