The authors show that in SR-BI knockout mice, plasma α-tocopherol is increased while α-tocopherol in several tissues is significantly decreased. The importance of this receptor in α-tocopherol uptake into tissues reveals that α-tocopherol by no means depends on passive trafficking between membranes alone, implicating the diversity of pathways that this molecule is dependent on in its metabolism.

A major enzymatic pathway of tocopherol biotransformation in mammalian tissues is described, involving the cytochrome P450 isoform 4F2 for initial ω-hydroxylation of both α- and γ-tocopherol. This was a major study showing that not only 'repackaging' of tocopherols in lipoproteins occurs in the body, but also metabolism to water-soluble metabolites play a role for effective excretion of tocopherols. The body seems to depend on a pathway to eliminate tocopherols effectively, emphasizing the possibility that tocopherols may have adverse effects if supplied too abundantly.
