Densitometry was accomplished with the NIH ImageJ software program. The amount of supercoiled DNA was multiplied by a factor of 1.22 to account for reduced ethidium bromide intercalation into the form I plasmid DNA.

Based on the reported limit of detection of DNA via photography of ethidium bromide stained gels (1 ng: see Tuma, R. S.; Beaudet, M. P.; Jin, X.; Jones, L. J.; Cheung, C.-Y.; Yue, S.; Singer V. L. Anal. Biochem. 1999, 268, 278), we are able to detect as little as 0.25% of our total DNA (400.2 ng in each reaction lane).
