The authors demonstrate that Saccharomyces cerevisiae γ-tubulin is phosphorylated and that a conserved tyrosine near the carboxyl terminus (Tyr445) is required for the phosphorylation. A mutation of this residue causes an increase in the frequency of new microtubules appearing in the spindle pole body region and this suggests that phosphorylation of γ-tubulin is important for regulating microtubule nucleation in Saccharomyces cerevisiae. Interestingly, mutation of this residue causes an arrest before anaphase and also alters the dynamics of the plus ends of microtubules.

Using RNA-mediated interference depletion of γ-tubulin in Caenorhabditis elegans embryos, the authors demonstrate that γ-tubulin is not absolutely required for microtubule nucleation. The phenotype conferred by the depletion of γ-tubulin suggests, however, that γ-tubulin is required for the normal organization and function of kinetochores and interpolar microtubules.
