Am J Hum Genet 1998, 62:824-833. WTI mutations, involving exons 8 and 9 and intron 9, have been identified in four out of 10 patients with isolated diffuse mesangial sclerosis. The absence of WT1 mutations in the other six patients suggests genetic heterogeneity of the disease.

Familial forms of FSGS have been identified, and linkage to chromosome 11q21-q22 was found in a large family whereas the disease was not linked to chromosome 11 in a second family neither to a previously described locus on chromosome 19. These results demonstrate genetic heterogeneity in familial FSGS.
