The crystal structure of native AdR was determined at 2.8 A resolution and that of the respective recombinant enzyme at 1.7 A resolution. The structure explains how the observed semiquinone is stabilised. It further shows that AdR has a highly asymmetric charge distribution, which most likely controls the approach of adrenodoxin, which previously has been shown to reveal an asymmetric charge distribution on its surface, too (see Müller [19]). A model of adrenodoxin/AdR interaction has been suggested, in which adrenodoxin binds to a positively charged cleft of the AdR.

Formation of an additional electrostatic interaction via Arg-418, in addition to participation of the conserved residues Lys-354 and Lys-358 in electrostatic contacts to adrenodoxin, has been identified as the reason for this observation. This indicates changes in adrenodoxin binding to different P450s.

It was demonstrated that truncated adrenodoxin mutants, possessing enhanced electron transfer abilities, produce increases in corticosterone and aldosterone biosynthesis when incubated with liposomal bovine CYP11B1. The effect of truncated adrenodoxin mutants on the product pattern of bovine CYP11B1 was confirmed in COS-1 cells. Moreover, it was demonstrated that in COS-1 cells the truncated mutants also enhance aldosterone formation relative to the formation of corticosterone by human CYP11B2. This implies that conditions changing the electron transfer abilities of adrenodoxin could have an impact on aldosterone formation, thus indicating a new regulatory level of aldosterone biosynthesis.

Leptin (0.6-18 nM) suppressed ovarian steroid synthesis costimulated by follicle-stimulating hormone and dexamethasone. Production of pregenenolone, progesterone, and 20α-hydroxy-4-pregnen-3-one was inhibited by leptin. The inhibition was due at least in part to reduced expression of adrenodoxin. This data indicates that changing the amount of adrenodoxin exerts a regulatory effect on adrenodoxin-dependent steroid biosynthesis.

EMBO J 1999, 18:5494-5504. It was demonstrated that CYP2B1 contains a chimeric mitochondrial and endoplasmic reticulum targeting signal at its N-terminus. Inducers of cyclic adenosine monophosphate and protein kinase A-mediated phosphorylation of CYP2B1 at Ser128 activate the signal for mitochondrial targeting and this way modulate its mitochondrial or endoplasmic reticulum destination. This observation demonstrates a novel regulatory mechanism by which cyclic adenosine monophosphate modulates the targeting of a protein to two distinct organelles.
