Horrobin et al reported that tetraacetate 8 was regioselective 6-deacetylation by the esterase-catalysed hydrolysis, gave the 6-hydroxy derivative 9 in 73% yield. We believed that treatment of compound 9 and benzyl succinate acid with DCC, 4-DMAP would afford compound 4 in excellent yield. The approach of enzymatic method offers a concise strategy to synthesize 4, it is unfortunate that the source of esterase (from R. toruloides) is not commerically available.
