J Biol Chem 1999; 274:1920-1927. A very careful study that analyzes the molecular mechanisms responsible for depressed Na-K-ATPase-mediated transport during dopamine stimulation. The activity and trafficking regulation of sodium pump alpha subunit are analyzed in parallel. The paper addresses the controversial issue of the role of phosphorylation of two serines found in the N-terminal region, and concludes that phosphorylation of one, not the other, mediates internalization, and that ATPase activity was unaffected by phosphorylation of either site.

Responses are strikingly similar to those seen with acute hypertension: internalization of apical NHE3 and NaPi and inhibition of Na-K-ATPase. It also dissects the signalling responsible for the responses by using PTH analogue, which does not stimulate AC.

This group was the first to demonstrate that microtubule-specrfic drugs could be used in the whole animal in vivo to analyze the role of microtubules in the trafficking of transporters. Using confocal microscopy, the paper very elegantly details the effects of the drugs on the spatial organization of the microtubules, as well as analyzes NaPi distribution biochemically and using microscopy.
