Scar1 and the related Wiskott-Aldrich syndrome protein, WASP, regulate the actin cytoskeleton through Arp2/3 complex
-
This is the first evidence for direct interaction between WASP-family proteins and the Arp2/3 complex. The authors identify the carboxy-terminal acidic domain as necessary for Arp2/3 binding and demonstrate that this domain acts as a dominant negative inhibitor of growth factor-induced actin reorganization in cultured cells.

WASP-related protein Scar activates dendritic nucleation of actin filaments by Arp2/3 complex
-
The authors demonstrate, for the first time, that WASP-family proteins stimulate the nucleation activity of the Arp2/3 complex and they show that activation requires both the Arp2/3-binding acidic and actin-binding WH2 domains. Full-length SCAR is less active than the C-terminal half of the molecule, suggesting that an N-terminal portion contains a regulatory domain. Also, actin filaments feed back positively on the activation of Arp2/3.

Arp2/3 complex and actin depolymerizing factor/cofilin in dendritic organization and treadmilling of actin filament array in lamellipodia

Arp2/3 localizes to the branch points between actin filaments at the leading edge of fish keratocytes. The beautiful electron micrographs in this paper are, perhaps, the best evidence to date that Arp2/3-mediated 'dendritic nucleation' of actin filament assembly is responsible for the structure of the leading edge of a motile cell.

Reconstitution of actin-based motility of Listeria and Shigella using pure proteins
-
This is the long-awaited reconstitution of L. monocytogenes motility from purified components. The most significant result is that motility requires only four components: actin, a nucleation and crosslinking factor (Arp2/3), a depolymerization factor (cofilin) and a barbed-end capping protein. Along with a spatially-localized activator of Arp2/3 (ActA) these proteins represent a minimal set of components required to construct a self-sustaining, actin-based motility engine.

Mechanism of Cdc42-induced actin polymerization in neutrophil extracts
-
Actin filaments nucleated in cell extracts by addition of activated Cdc42 appear to be capped more slowly than exogenous actin seeds added to the extract. The results suggest that Cdc42-nucleated filaments are maintained in an uncapped state, possibly by an elongation-promoting factor associated with the growing ends of the filaments.

Zigmond S.H., Joyce M., Yang C., Brown K., Huang M., Pring M. Mechanism of Cdc42-induced actin polymerization in neutrophil extracts. J Cell Biol. 142:1998;1001-1012.

G protein signaling events are activated at the leading edge of chemotactic cells
-
The activity of the signaling system responsible for eukaryotic chemotaxis is spatially polarized. The results suggest that cell polarity can be established in the absence of an assembled cytoskeleton.
