The three-dimensional structure of murine IL-11 has been resolved. The molecule has a basic 4 alpha bundle structure similar to several other cytokines and growth factors. There are three major contact points with its cellular receptor: the IL-11 alpha receptor site, the primary GP130 binding site, and a secondary GP130 receptor binding site. It is assumed that the human IL-11 structural interactions with its receptor are similar to the murine system. Confirmation of the human IL-11: IL-11Rα/GP130 receptor interaction awaits crystallization and analysis of the three-dimensional structure human IL-11.

Interleukin-11 is not necessary for normal hematopoiesis but it is absolutely essential for normal fertility.

Neutrophils are not affected by the anti-inflammatory actions of IL-11 because the IL-11 alpha receptor is not expressed on the cellular surface of polymorphonuclear cells. In this respect, IL-11 differs from IL-10, which will downregulate neutrophil expression of pro-inflammatory cytokines.

Using a transgenic mouse model with targeted over-expression of IL-11 within lung tissue, these investigators were able to demonstrate the epithelial protective effects of IL-11 after oxidant injury to pulmonary tissues. The findings indicate that the protective effects of IL-11 against damage to gastrointestinal epithelial tissues extend to the pulmonary mucous membranes as well.

J Pediatr Surg 1998, 33:24-29. After massive small bowel resection in a rat model, IL-11 had additive effects on small bowel growth potential with epidermal growth factor. Epidermal growth factor lead to generalized enterocyte and myocyte growth, whereas IL-11 acted upon enterocytes only. Interleukin-11 may prove to be efficacious in the clinical management of short bowel syndrome in patients after extensive small bowel resection.

This study examines the therapeutic value of a combination of granulocyte colony-stimulating factor with IL-11 in an experimental model of Pseudomonas aeruginosa sepsis in the neutropenic rat. The combination of G-CSF and IL-11 was significantly more effective than either cytokine given alone (P<0.001) in reducing the mortality rate from septic shock, and IL-11 (but not G-CSF) protected the gut epithelium from damage after the chemotherapy-induced period of neutropenia.
