2 Keith Jr JC: Interleukin-11. In The cytokine reference: a compendium of cytokines and other mediators of the host defense. Edited by Oppenheim J, Feldman M. San Diego: Academic Press 1999, 6:1-58. This is a definitive review of the entire published database regarding interleukin-11 up to the present. The paper is a part of an online publication that reviews all the available literature on the cytokines. This is an excellent reference source on IL-11 biology.

Identification of three distinct receptor binding sites for murine interleukin-11
-
5 Barton VA, Hudson KR, Heath JK: Identification of three distinct receptor binding sites for murine interleukin-11. J Biol Chem 1999, 274:5755-5761. The three-dimensional structure of murine IL-11 has been resolved. The molecule has a basic 4 alpha bundle structure similar to several other cytokines and growth factors. There are three major contact points with its cellular receptor: the IL-11 alpha receptor site, the primary GP130 binding site, and a secondary GP130 receptor binding site. It is assumed that the human IL-11 structural interactions with its receptor are similar to the murine system. Confirmation of the human IL-11: IL-11Rα/GP130 receptor interaction awaits crystallization and analysis of the three-dimensional structure human IL-11.

Recombinant human interleukin-11 does not affect functions of purified human neutrophils in vitro
-
9 Bozza N, Kyvelos D, Trepicchio WL, Collins M, Klempner NS, Dorner AJ: Recombinant human interleukin-11 does not affect functions of purified human neutrophils in vitro. J Interferon Cytokine Res 1998, 18:889-895. Neutrophils are not affected by the anti-inflammatory actions of IL-11 because the IL-11 alpha receptor is not expressed on the cellular surface of polymorphonuclear cells. In this respect, IL-11 differs from IL-10, which will downregulate neutrophil expression of pro-inflammatory cytokines.

Interleukin-11 effects in acute graft-vs.-host-disease: Role of Th2 polarization, gastrointestinal protection and inflammatory cytokine inhibition

J Clin Invest 1998, 102:115-123. Interleukin-11 was highly beneficial in a murine model of graft versus host disease after allogeneic bone marrow transplantation. Interleukin-11 prevented mucosal injury from graft versus host disease, reduced systemic endotoxin levels, promoted Th2 polarization, and attenuated interferon-γ levels after bone marrow transplant.

Targeted lung expression of interleukin-11 enhances murine tolerance of 100% oxygen and diminishes hyperoxia-induced DNA fragmentation
-
18 Waxman AB, Einarsson O, Seres T, Knickelbein RG, Homer R, Warshaw JB, Johnston R, Elias JA: Targeted lung expression of interleukin-11 enhances murine tolerance of 100% oxygen and diminishes hyperoxia-induced DNA fragmentation. Chest 1999, 86:340-348. Using a transgenic mouse model with targeted over-expression of IL-11 within lung tissue, these investigators were able to demonstrate the epithelial protective effects of IL-11 after oxidant injury to pulmonary tissues. The findings indicate that the protective effects of IL-11 against damage to gastrointestinal epithelial tissues extend to the pulmonary mucous membranes as well.

J Pediatr Surg 1998, 33:24-29. After massive small bowel resection in a rat model, IL-11 had additive effects on small bowel growth potential with epidermal growth factor. Epidermal growth factor lead to generalized enterocyte and myocyte growth, whereas IL-11 acted upon enterocytes only. Interleukin-11 may prove to be efficacious in the clinical management of short bowel syndrome in patients after extensive small bowel resection.

24 Peterson RL, Wang L, Keith JC Jr, Dorner AJ: Molecular effects of recombinant human interleukin-11 in the HLA-B27 rat model of inflammatory bowel disease. Lab Invest 1998, 78:1503-1512. In this transgenic rat model that mimics human inflammatory disease, IL-11 led to decreased disease activity and reduced colonic RNA expression of TNF, IL-1, and interferon-γ. Blood 1999, 93:3467-3472. This study examines the therapeutic value of a combination of granulocyte colony-stimulating factor with IL-11 in an experimental model of Pseudomonas aeruginosa sepsis in the neutropenic rat. The combination of G-CSF and IL-11 was significantly more effective than either cytokine given alone (P<0.001) in reducing the mortality rate from septic shock, and IL-11 (but not G-CSF) protected the gut epithelium from damage after the chemotherapy-induced period of neutropenia.
