    • Vesicular tubular clusters between the ER and Golgi mediate concentration of soluble secretory proteins by exclusion from COPI-coated vesicles
    • This quantitative immunoelectron microscopic study of endogenous proteins shows that secretory proteins of the exocrine pancreas leave the ER without prior concentration and reveals a role for VTC in cargo concentration by exclusion from retrograde COPI-vesicles. It also, for the first time, shows concentration of an endogenous membrane protein at the COPII-coated ER exit sites, suggesting that proteins may exit the ER either selective or by bulk flow.

    • Mutation of the DXE code in the VSV-G tail unexpectedly reveals that the recruitment of VSV-G to prebudding complexes is not altered but the subsequent concentration step is lacking. These data imply that concentration of some proteins at the ER exit sites is achieved by selective retention in COPII vesicles rather than by active recruitment.
