    • This paper reports the cloning of EIN2, which plays a central role in ethylene signaling. The amino-terminal portion of the predicted EIN2 protein contains 12 transmembrane helices with similarity to metal ion transporters. The carboxy-terminal portion is a novel hydrophilic domain that confers ethylene responses when overproduced in the ein2 mutant background.

Receptor mutations, which are known to prevent ethylene binding, also prevent binding of copper.     • Previously, the only mutations that had been isolated for any ethylene receptor genes were dominant mutations conferring ethylene insensitivity. In this paper, null (or severe loss-of-function) mutations were isolated for four of the five Arabidopsis ethylene receptor genes. Phenotypic analyses indicate that these genes are at least partially redundant since individual mutants appear wild type. Furthermore, the genes are negative regulators of ethylene responses, because when crossed together to create triple or quadruple mutants, constitutive ethylene responses are obtained.

Either gene confers ethylene insensitivity to Arabidopisis when the mutation known to block ethylene binding in Arabidopsis etr1-1 is introduced into them.     • The relationship between ethylene binding and dominant insensitivity conferred by mutant forms of the ETR1 ethylene receptor
    • in press. The effect of ETR1 dominant mutations on ethylene binding is examined in order to better understand how these alleles confer ethylene insensitivity. Mutations that lie in the first and second hydrophobic subdomains of ETR1 prevent ethylene binding, whereas a mutation in the third hydrophobic subdomain does not. The latter mutation is thought to uncouple ETR1 output activity from ethylene binding.

Plant Physiol. 1999;. in press. The effect of ETR1 dominant mutations on ethylene binding is examined in order to better understand how these alleles confer ethylene insensitivity. Mutations that lie in the first and second hydrophobic subdomains of ETR1 prevent ethylene binding, whereas a mutation in the third hydrophobic subdomain does not. The latter mutation is thought to uncouple ETR1 output activity from ethylene binding.
