Selection of mutants of these transporters expressed in S. pombe deficient in deoxyglucose uptake allowed identification of residues involved in exofacial binding. Site-directed mutagenesis experiments pinpointed the residues involved in D-galactose selectivity in one of the HUP transporters.

This article describes an integrated model of lac permease structure and function, based mainly on data from cysteine-scanning mutagenesis. The authors provide a rational model for both orientation of the 12 membrane-spanning helices and a detailed molecular picture of the substrate translocation event coupled to a proton-motive force.

This study reports the striking observation that random mutagenesis of hexose transporters in yeast can confer on these proteins the ability to mediate the transport of cations. The mutations observed were found to cluster into regions of the transporters that were previously identified as functionally important for hexose uptake (and some additional sites) and suggest that the substrate permeation pathway for major facilitator superfamily members might be relatively nonspecific in certain circumstances.

Biochemistry 1998, 37:1322-1326. This study identified a conserved QLS (Gln-Leu-Ser) sequence within the glucose-specific transporters, which discriminates against D-fructose selection at the exofacial binding-site. Substitution of the QLS motif in Glut3 with the corresponding HVA (His-Val-Ala) sequence from Glut2 resulted in a gain-of-function, such that Glut3 was capable of some D-fructose transport.

This chimeric transporter study demonstrates that Glut2 contains at least two crucial domains, which separately modulate the recognition of D-fructose (helices 7 and 8) and the kinetic properties of D-glucose transport (helices 9-12). These authors propose a functional interaction between the carboxyl terminus of Glut2 and such regions.
