Impact of preexisting and induced humoral and cellular immune responses in an adenovirus-based gene therapy phase I clinical trial for localized mesothelioma

Hum Gene Ther 1998, 9:2121-2133. Little is known about the immune response in humans to adenovirus gene therapy vectors. This paper shows that out of 21 patients, most had preexisting neutralizing antibodies and had peripheral blood mononuclear cells that proliferated in response to adenovirus. These antibodies did not preclude transgene (herpes simplex virus thymidine kinase) expression. Vector instillation induced high titers of neutralizing antibodies against adenovirus.

This paper identifies a new protein, FLASH, that is a component of the DISC. FLASH contains a death effector domain (DED) like sequence that interacts with the DED of Procaspase 8 and it contains a CED-4/Apaf-1 like sequence that mediates oligomerization. FLASH may mediate the ability of Fas or FADD to activate Procaspase 8. FLASH interacts with E1B-19K and overcomes its ability to inhibit Fas induced apoptosis. The work provides further insight into the mechanism of action of E1B-19K.

The work therefore proves that E3-14.7K has anti-inflammatory properties in vivo and it underscores the possibility of using E3-14.7K in gene therapy.
