Hum Gene Ther 1998, 9:2121-2133. Little is known about the immune response in humans to adenovirus gene therapy vectors. This paper shows that out of 21 patients, most had preexisting neutralizing antibodies and had peripheral blood mononuclear cells that proliferated in response to adenovirus. These antibodies did not preclude transgene (herpes simplex virus thymidine kinase) expression. Vector instillation induced high titers of neutralizing antibodies against adenovirus.

    • Imai Y, Kimura T, Murakami A, Yajima N, Sakamaki K, Yonehara S: The CED-4-homologous protein FLASH is involved in Fas-mediated activation of caspase-8 during apoptosis. Nature 1999, 398:777-785. This paper identifies a new protein, FLASH, that is a component of the DISC. FLASH contains a death effector domain (DED) like sequence that interacts with the DED of Procaspase 8 and it contains a CED-4/Apaf-1 like sequence that mediates oligomerization. FLASH may mediate the ability of Fas or FADD to activate Procaspase 8. FLASH interacts with E1B-19K and overcomes its ability to inhibit Fas induced apoptosis. The work provides further insight into the mechanism of action of E1B-19K.

Immunology 1999, 96:1042-1047. This paper identifies a new and interesting protein, FIP-3, that binds to E3-14.7K and plays a role in TNF signal transduction - leading to apoptosis and activation of NFκB. FIP-3 binds to RIP and NIK. The work provides a major insight into how E3-14.7K may function.

    • Perez D, White E: E1B 19K inhibits Fas-mediated apoptosis through FADD-dependent sequestration of FLICE. J Cell Biol 1998, 141:1255-1266. E1B-19K is known to be a functional homolog of BCL-2. This paper shows that E1B-19K can also inhibit Fas induced apoptosis at another level, namely via FADD and Procaspase 8 in the DISC.

    • Lung-specific expression of E3-14.7K in transgenic mice attenuates adenoviral vector-mediated lung inflammation and enhances transgene expression
    • Harrod KS, Hermiston TW, Trapnell BC, Wold WSM, Whitsett JA: Lung-specific expression of E3-14.7K in transgenic mice attenuates adenoviral vector-mediated lung inflammation and enhances transgene expression. Hum Gene Ther 1998, 9:1885-1898. This paper shows that E3-14.7K expressed in the lungs of transgenic mice strongly inhibits the inflammatory and pathogenic response to adenovirus. The work therefore proves that E3-14.7K has anti-inflammatory properties in vivo and it underscores the possibility of using E3-14.7K in gene therapy.
