    • The first report on the requirement for the en and hkb transcription factors in the development of serotonergic neurons in Drosophila. Having established that the serotonergic neurons of the VNC are derived from NB7-3, which expresses en and hkb, the authors show that loss-of-function mutations in either gene lead to a severe reduction in the number of 5HT-expressing neurons in the trunk. They also show that the serotonergic neurons in the brain are preserved.

Conversely, ectopic expression of eg produces extra serotonergic neurons. This ectopic expression of 5HT depends on hkb, suggesting a combinatorial action of the two transcription factors on the serotonergic phenotype. The data also show that eg is required for correct axonal pathfinding in the NB7-3 lineage.

    • A comprehensive study on the Drosophila homologue of the vertebrate Islet1 and Islet2 genes, shown to be expressed early on in non-neural tissues, and then in a subset of motor neurons and in the dopaminergic and serotonergic neurons of the VNC. islet loss-of-function mutations in Drosophila result in axonal pathfinding errors by the three neuronal types that normally express the gene, and in the loss of TH and 5HT expression in the VNC. Ectopic expression generates ectopic TH-expressing cells, but not 5HT-expressing ones, and causes alterations in motor neuron pathfinding.

    • Alternative neural crest cell fates are instructively promoted by TGFbeta superfamily members
    • An important in vitro study on fate determination of neural crest cells by extracellular factors showing that BMP2 and TGFβ1 promote neuronal and smooth muscle differentiation, respectively. In the context of this review, the most interesting results are that BMP2 induces expression of Mash1 and is present at the right time and place to act on sympathetic precursors.

    • Involvement of bone morphogenetic protein-4 and bone morphogenetic protein-7 in the differentiation of the adrenergic phenotype in developing sympathetic neurons
    • Demonstrates that BMP4 and BMP7, shown to be expressed in vivo where and when sympathetic precursors develop, induce TH (and Phox2a) in neural crest cultures (see also [43,44]). In addition, the authors present the first in vivo gain-of-function data showing that overexpression of BMP4 results in enlargement of sympathetic ganglion primordia and the presence of ectopic TH-positive cells.

    • Specification of motor neuron identity by the MNR2 homeodomain protein
    • Discovered by ingenious use of single-cell reverse transcription (RT)-PCR technology, the chicken homeodomain transcription factor MNR2 is shown to be specific for progenitors and early postmitotic precursors of somatic motor neurons. The authors also show that retrovirus-mediated ectopic expression of MNR2 in the neural tube induces transcription factors shown, or supposed, to be involved in motor neuron differentiation. In the context of this review, it is important to note that MNR2 is sufficient to induce ChAT and to re-direct axonal trajectories. It remains to be seen whether MNR2 itself or, what seems more likely, one of the downstream transcription factors (and which one) activates the ChAT gene directly.
