Intracellular cytokine profile of cord and adult blood lymphocytes
-
1 Chalmers IM, Janossy G, Contreras M, Navarrete C: Intracellular cytokine profile of cord and adult blood lymphocytes. Blood 1998, 92:11-18. The cytokine profile of cord blood lymphocytes assessed at the single-cell level showed a reduced production of IL-2, IL-4, IFN-gamma, and TNF-α, as compared to adult peripheral blood lymphocytes. The authors suggest that the reduced incidence of acute graft-versus-host disease in cord blood transplantation may partly due to the altered cytokine profile seen in cord blood.

A comparison of cord blood and adult blood cytokine production by stimulated mononuclear cells showed a reduced production of IFN-gamma, TNF-α, and IL-4 by cord blood cells. The authors suggest that the cytokine production pattern that characterizes cord blood could provide an explanation for the lower occurrence of acute graft-versus-host disease following cord blood transplant.

Pediatr Res 1998, 44:469-477. An evaluation of cytokine plasma levels and gene expression in healthy neonates and in infants with clinically suspected or confirmed sepsis is presented in this paper. The results suggest that cord plasma levels of G-CSF, tumor necrosis factor-α, interleukin-1β, interleukin-6, and interleukin-8 can predict early-onset neonatal sepsis with high sensitivity and specificity.

This review of cord blood cytokine production and function is reported by investigators who have contributed much to the knowledge of neonatal hematopoiesis and immune function over the past decade. It discusses the mechanisms associated with the dysregulation of GM-CSF, M-CSF, interleukin-12, and interleukin-15; the effect of these cytokines on induction of interferon gamma and tumor necrosis factor-α production; and their effect on natural killer cell and lymphokine-activated killer cell activities in cord blood. The authors conclude that the reduced cytokine expression from activated cord blood may contribute to the impaired neonatal cellular immunity and suggest that exogenous cytokines may compensate for the immaturity of cord blood cells.

Although no significant differences were found between normal term and preterm infants, no response to thrombocytopenia was observed in preterm infants, suggesting that impaired fetal thrombopoietin production may be a factor in early thrombocytopenia in preterm infants.
