Hox genes in brachiopods and priapulids and protostome evolution

This paper demonstrates that invertebrate Hox genes support a division of the protostomes into two major clades, one of which comprises arthropods, nematodes and priapulids, which share a specific posterior Hox gene, the other comprising annelids, nemerteans, gastropods, flatworms, and brachiopods, all of which share another, different species of posterior Hox gene. A subset of anterior Hox genes shared by the two protostome clades and the deuterostomes suggests that the bilateralian ancestor to these three clades possessed at least eight Hox genes. These findings are consistent with ribosomal DNA sequence analyses that suggested the existence of a singular deuterostome clade and two divergent clades of protostomes.

Although Amphioxus possesses a rostral cerebral vesicle, it lacks an anatomically partitioned forebrain. However, Amphioxus shares with chordates and ascidians Otx expression. Did the craniate descendants of a cephalochordate-like ancestor (such as the Cambrian taxa Yunanozoon or Pikaia) evolve olfactory forebrain neuropils from an ancient cohort of rostral organizer genes? The studies of Williams and Holland support this notion. They propose that compartmentalization of the neural tube in ascidians into three components (equivalent to the chordate fore- and midbrain, isthmus, and hindbrain) by rostral expression domains of a suite of genes (i.e. the Otx homologue, Hroth; the Pax-6 homologue, HrPax-258; and the HrHox1 gene) is ancient and has been carried over into the chordates. In chordates, evolution of increased complexity from ascidian-Amphioxus type organization may be a consequence of gene duplication and clustering, which provided novel developmental interactions for novel forebrain partitions. (see also [71]) This would suggest de novo evolution of olfactory forebrain and a glomerular olfactory lobe in the craniates.

This comparative study confirms and extends the view that the superfamily of vertebrate olfactory-receptor genes is divisible into two classes - classes I and II - whose receptor proteins may recognize water-soluble and airborne odorants, respectively. Teleost fish have class I receptor genes, terrestrial mammals have class II receptor genes, and amphibians have both. The coelocanth Latimeria chalumnae has both classes, but most of its class II receptor genes are non-functional pseudogenes. By contrast, a marine mammal, the dolphin Stenella coeruleoalba, has only class II receptor genes, all of which were found to be non-functional pseudogenes.
