    • ComP is the membrane sensor for the ComX pheromone. PhoA and LacZ fusions demonstrate that the amino-terminal membrane-localized moiety of ComP has six or eight transmembrane segments, as well as two large extracellular loops. Deletion of these loops confers ComX independence on ComP function.

    • Chemical synthesis confirms that the agr system pheromone contains a thiolactone necessary for biological activity. Analysis of pheromone analogs demonstrate that the activating and inhibiting activities depend on different chemical properties and are therefore due to distinct receptor interactions. It is proposed that the activating interaction involves a transacylation reaction in which the thiolactone is attacked by a nucleophile on the receptor and that the inhibition reaction involves a non-covalent interaction that blocks binding of the cognate pheromone.
