    • 3H)-KYN (10 Ci/mmol) and 3 μl of different concentration of inhibitor solutions. After the incubation, the reaction was terminated by addition of 300 μl of 7.5% (w/v) activated charcoal, and centrifuged for 7 min. A 75μl aliquot of supernatant was transferred to optiplate and liquid scintillation added. The optiplates were vortexed and the radioactivity counted in a microplate scintillation counter. The apparent discrepancies in potency as compared to the rat brain enzyme seem to be due to the lesser sensitivity of the radiometric detection rather than to different tissue specificity.

    • 4. After centifugation, supernatants were directly used for the measurement of 3,4 dihydroxybenzoic acid. The concentration of 3,4 dihydroxybenzoic acid was measured by HPLC with coulometric detection (det. 1 +0.05V, det 2 +0.2V) using a 10 cm C18 reversed phase (3μm) column with a mobile phase containing water / acetonitrile / trifluoroacetic acid (95 / 5 / 0.05). The flow rate was 1 ml/min.
