This study is important for two reasons. First, the investigators used clinically relevant UHMWPE wear debris and developed osteolysis around uncemented total hip replacements in a canine model. Second, in another group of canines with similar hip replacements and introduction of wear debris, an oral treatment with bisphosphonates could inhibit localized periprosthetic bone loss.

In this study the investigators demonstrated that clodronate, a first-generation bisphosphonate, can be toxic to macrophages. This effect was specific to individual drug formulations, and alendronate had no deleterious effect.
