Until recently histidine kinase had only been found in bacteria, but work in the past few years uncovered a set of related proteins in eukaryotes. The shared characteristics of eukaryotic histidine kinases show that they are all hybrid kinases that play a variety of different physiological roles.

Several Dictyostelium genes were discovered among the products generated by PCR using primers designed on the basis of established histidine kinases. Cells lacking DhkB were found to grow and develop normally but to make spores that germinated prematurely. A secreted adenine derivative is responsible for dormancy of spores while they remain on top of fruiting bodies and its effects may be mediated by DhkB.

Microbiology 1998, 144:425-432. The yeast Candida albicans was found to carry close homologs of both SLN1 of Saccharomyces cerevisiae and NIK1 of Neurospora crassa, which are involved in filamentous growth. Because C. albicans is the cause of life-threatening infections in immunocompromised patients, these histidine kinases present attractive targets for new drugs.

The convergence of classic and molecular genetics confirms the phenotype resulting from loss of this histidine kinase.

A mutant isolated by random restricted enzyme mediated integration (REMI) mutagenesis develops more rapidly than wild-type cells. The disrupted gene, rdeA, encodes a small protein with a histidine motif related to that of the yeast phosphorelay protein, Ypd1 p. Null mutations in the rdeA gene can be complemented by the yeast YPD1 gene, indicating that their products may play similar roles.

Genetic studies had shown that the HOG (high osmolarity glycerol response) mitogen-activated protein (MAP) kinase cascade leading to osmotic resistance of yeast is activated following a shift to high osmotic conditions. Under standard osmotic conditions Sln1p is active and relays a phosphate to Ssk1p that inhibits its interaction with the MAP kinase kinase kinases, Ssk2p and Ssk22p. When Ssk1p no longer receives a phosphate from Sln1p following a shift to high osmolarity, it interacts with Ssk2p and stimulates its autophosphorylation. The MAP kinase pathway remains active even after Ssk1 p is rephosphorylated, because of the autophosphorylation of Ssk2p.

A second osmosensor in the yeast Saccharomyces cerevisiae, Sho1p, activates the mitogen-activated protein kinase kinase kinase (MAPKKK), Ste11p, that is bound by the MAPK kinase Pbs2p. This route parallels that by which Slnip activates the MAPK kinase, Ssk2p, but leads to the same destination, the high osmolarity glycerol response (HOG1) MAPK. Ste11p also acts in the pheromone signal transduction pathway where it is held to Ste7p (MAPK kinase) and Fus3p (MAPK) by the scaffold protein Ste5p.
