By examining the incorporation of [35S]methionine into 43S preinitiation complexes as isolated on CsCI density grardients, a direct effect of sepsis on one of the early steps in peptide-chain initiation was demonstrated. These studies therefore identify one biochemical locus responsible the inhibition of protein synthesis in sepsis.

Am J Physiol 1996; 270:E43-E50. These studies showed that sepsis, but not sterile inflammtion cause expression of elF-2B protein to be diminished. The decrease in protein paralleled a similar reduction in the mRNA suggesting that the protein content is regulated by mRNA expression. No alteration in elF-2B activity was observed in heart, a tissue where protein synthesis is not affected by sepsis. The depletion of elF-2B protein provides in part a biochemical reason for reduced elF-2B activity in fast-twitch muscle during sepsis.

By using a specific inhibitor of IL-1, the authors were able to demonstrate that prevention of the fall in protein synthesis with this compound was attributed to a maintanence of elF-2B protein and mRNA at levels observed in control individuals.
