Inherited susceptibility determines the distribution of dense low density lipoprotein subtraction profiles in familial combined hyperlipidemia
-
Bredie SJH, Kiemeney LA, de Haan AFJ, Demacker PNM, Stalenhoef AFH. Inherited susceptibility determines the distribution of dense low density lipoprotein subtraction profiles in familial combined hyperlipidemia. Am J Hum Genet 1996; 58:812-822. In this study the inheritance of small, dense LDL subfraction profiles, based on LDL particles density, was investigated in 40 well defined Dutch FCH families. A major autosomal locus recessively inherited, with a high population frequency, and an additional small polygenic component best explained the LDL subfraction distribution in these families.

Arterioscler Thromb Vasc Biol 1997; 17:748-754. Impaired insulin action in FCH patients was demonstrated, both on the suppression of nonesterified fatty acids and on stimulation of glucose disposal. A primary defect in fatty acid metabolism may contribute to the lipid abnormalities in this disorder.

Lovastatin decreases de novo cholesterol synthesis and LDL apoB-100 production rates in combined-hyperlipidemic males
-
Cuchel M, Schaefer EJ, Millar JS, Jones PJH, Dolnikowski GG, Vergani C, Lichtenstein AH. Lovastatin decreases de novo cholesterol synthesis and LDL apoB-100 production rates in combined-hyperlipidemic males. Arterioscler Thromb Vasc Biol 1997; 17:1910-1917. This stable isotope kinetic study investigated the effect of lovastatin on apoB metabolism in male FCH patients. The decline in plasma lipid levels after treatment with lovastatin was attributable to reductions in the production rate and fractional synthesis rate of de-novo cholesterol synthesis and the production rate of apoB-100-containing lipoproteins. A comprehensive review on published data on variations in metabolism of apoB-containing lipoproteins is included.

The redox status of coenzyme Q10 in total LDL as an indicator of in vivo oxidative modification; studies on subjects with familial combined hyperlipidemia
-
de Rijke YB, Bredie SJH, Demacker PNM, Vogelaar JM, Hak-Lemmers HLM, Stalenhoef AFH. The redox status of coenzyme Q10 in total LDL as an indicator of in vivo oxidative modification; studies on subjects with familial combined hyperlipidemia. Arterioscler Thromb Vasc Biol 1997; 17:127-133. Total LDL in FCH patients was less well protected against in-vitro oxidation than was total LDL from normolipidaemic relatives. In addition, the decreased redox status of coenzyme Q10 in LDL from individuals with FCH suggests that the LDL in the circulation is already modified because of lipid peroxidation.
