In this study a new method of FISH is described in which DNA probes that span the common breakpoints of the Philadelphia chromosome are used, which detect double fusions in true positives, thereby decreasing the false positive rate and increasing the sensitivity of MRD detection.

Detectable molecular residual disease at the beginning of maintenance therapy indicates a poor outcome in children with T-cell acute lymphoblastic leukemia. Blood 1997, 90:1226-1232, An important study that uses TCR rearrangements to detect MRD. In this study it was observed that the presence of MRD was a strong predictor of a poor outcome at several distinct time points. Results from MRD assays performed in this manner may allow for the appropriate aggressive treatment of patients who are at high risk of relapse.

Autologous bone marrow transplantation for acute promyelocytic leukemia in second remission: prognostic relevance of pretransplant minimal residual disease assessment by reverse-transcription polymerase chain reaction of PML/RAR alpha fusion gene. Blood 1997, 90:1321-1325, In this study of 15 patients with the M3 subtype of acute myelocytic leukemia, the finding of PCR-positive disease prior to autografting strongly predicted for a poor outcome following an autologous transplantation in second complete remission. In contrast, those patients in second complete remission who were PCR negative prior to transplantation fared well.
