The development and maintenance of postobstructive polyuria in bilateral ureteral obstruction and release of this blockade are associated with a marked reduction in AQP2 expression in rat collecting ducts.

    • Congestive heart failure in rats is associated with increased expression and targeting of aquaporin-2 water channel in collecting duct
    • Nielsen S, Terris J, Andersen D, Ecelbarger C, Frokiaer J, Jonassen T, et al.: Congestive heart failure in rats is associated with increased expression and targeting of aquaporin-2 water channel in collecting duct. Proc Natl Acad Sci U S A 1997, 94:5450-5455. The authors demonstrate that the increased water retention in rats with congestive heart failure coincides with a marked increase in AQP2 expression and that nearly all AQP2 is located in the apical membrane of collecting duct cells. • Xu DL, Martin PY, Ohara M, St John J, Pattison T, Meng X, et al.: Upregulation of aquaporin-2 water channel expression in chronic heart failure. J Clin Invest 1997, 99:1500-1505. The increased water retention in rats with congestive heart failure coincides with a marked increase in AQP2 expression, and nearly all AQP2 is located in the apical membrane of collecting duct cells. In addition, administration of a vasopressin type 2 receptor antagonist, OPC31260, inhibited this up-regulation completely and resulted in a significant increase in diuresis, a decrease in urinary osmolality, and a rise in plasma osmolality. J Clin Invest 1997, 100:957-962. The generation of AQP4-knockout mice is reported. These mice have a mild defect in urinary concentrating ability consistent with the expression of AQP4 in the medullary collecting duct. The AQP4 deletion in these mice has little or no effect on development, survival, growth, and neuromuscular function. In addition, AQP4-knockout mice are free of symptoms related to disturbances in centra osmoregulation.
