Syntaxin-4 is localized to the apical plasma membrane of rat renal collecting duct cells: Possible role in aquaporin-2 trafficking
-
Mandon B, Chou CL, Nielsen S, Knepper MA. Syntaxin-4 is localized to the apical plasma membrane of rat renal collecting duct cells: possible role in aquaporin-2 trafficking. J Clin Invest 1996; 98:906-913. In this study, it was shown that syntaxin-4 is present in the apical membrane of AQP2-expressing collecting duct cells. Because a complementary protein (VAMP-2 [8,9]) in vesicle docking and fusion machinery has previously been identified in AQP2-containing vesicles of the same cells, it is suggested that these proteins are involved in fusion of AQP2-containing vesicles with the apical membrane of collecting duct cells.

Apical extracellular calcium/polyvalent cation-sensing receptor regulates vasopressin-elicited water permeability in rat kidney inner medullary collecting duct. J Clin Invest 1997; 99:1399-1405. Aquaporin-2 transfection of MDCK cells reconstitutes vasopressin-regulated transcellular osmotic water transport
-
Deen PMT, Rijss JPL, Mulders SM, Errington RJ, Van Baal J, Van Os CH. Aquaporin-2 transfection of MDCK cells reconstitutes vasopressin-regulated transcellular osmotic water transport. J Am Soc Nephrol 1997; 8:1493-1501. In this study it was shown that in an AQP2-transfected cell line the vasopressin-induced increase in transcellular osmotic water permeability coincides with the translocation of AQP2 from intracellular vesicles to the apical membrane of the cells. These cells are a good model to study short-term regulation of AQP2.

J Biol Chem 1996; 271:24365-24370. In this study it was shown the first time that in a transfected human collecting duct cell AQP2 is translocated from intracellular vesicles to the apical membrane after incubation with vasopressin, which increases the water permeability of the apical membrane sixfold. These cells provide a good model to study short-term regulation of AQP2.

Role of renal aquaporins in escape from vasopressin-induced antidiuresis in rat
-
Ecelbarger CA, Nielsen S, Olson BR, Murase T, Baker EA, Knepper MA, et al. Role of renal aquaporins in escape from vasopressin-induced antidiuresis in rat. J Clin Invest 1997; 99:1852-1863. In this study it was shown that in rats the increase in water excretion after a water load despite sustained administration of vasopressin is associated with a marked reduction of AQP2 protein and mRNA levels. This result suggests that this 'escape' from vasopressin-induced antidiureses is, at least in part, attributable to a vasopressin-independent decrease in AQP2 expression levels.

In this study it was shown for the first time that (puromycin aminonucleoside-induced) nephrosis is associated with a marked reduction in AQP2 expression in rat collecting ducts, despite increased circulating vasopressin.
