    • Bowerman B: Maternal control of pattern formation in early Caenorhabditis elegans embryos. Curr Top Dev Biol 1998, 39:73-117. Intensive genetic screening efforts in several laboratories have led to the identification of a large number of maternal genes responsible for early embryonic development in C. elegans. This review covers the current understanding of the maternal factors that have since been molecularly identified and their role in asymmetric cell division during early embryogenesis and in determining the identity of early blastomeres. An effort is made to categorize the maternal genes into three groups on the basis of their functions.

The authors of this paper describe the method of RNA-mediated interference in C. elegans, a technique that now makes it possible to rapidly assess the in vivo function of cloned gene. It is shown that antisense, sense, and (in particular) double-stranded RNAs are able to inhibit gene function in offspring when injected into mothers, remarkably even when RNA is injected in somatic tissue of the mother. The study indicates that only a few molecules of RNA are required for an effect and suggests that inhibition of gene activity is occurring at the level of the gene as opposed to the mRNA level.

Genes Dev 1997, 11:2883-2896. A description of the first identification of a zygotic gene, end-1, required to specify the identity of a founder cell, the endoderm progenitor or E cell. In a large deletion that removes this gene, E is converted to a C-like founder cell; end-1 restores the normal E cell lineage pattern to such mutants. end-1 encodes a GATA factor whose message is expressed specifically in the early E lineage, suggesting that endoderm is specified by a molecularly conserved mechanism in worms, flies, and vertebrates.

Development 1998, 125:2171-2180. This paper shows that pha-4, a gene required for formation of the pharynx and rectum in C. elegans, is a homologue of Drosophila fork head and mammalian HNF-3. The PHA-4 protein is expressed in all pharynx nuclei and binds to a regulatory region of the pharynx myosin-encoding myo-2 gene. The overall expression pattern is shown to be similar to that of fork head - high in foregut (pharynx) and hindgut (rectum) and low in the endoderm or midgut (intestine), suggesting a conserved mechanism for specification of the digestive tract across metazoa.

It is shown that a pha-4 mutation transforms pharynx cells into ectoderm and ectopic pha-4 leads to overproduction of pharynx-specific cells. PHA-4 is expressed throughout the pharynx and in the midgut (endoderm) and hindgut (rectum), suggesting conservation of the pathways that specify the animal digestive system.
