This paper finally settles the important, but difficult to address, question of how the C. elegans embryo sets A-P polarity. By using mutant backgrounds that increase the variability of the sperm entry point, the authors were able to determine that embryonic polarity correlates with the position of the sperm and not with any inherent polarity within the oocyte. The authors went on to show that the early cytoplasmic reorganization that accompanies the establishment of polarity ia always directed toward the sperm and that this relationship holds true in a number of related nematode species. These results focus future attention on the role of the sperm and on post-fertilization events, especially upon cytoplasmic reorganization.

The results focus attention on phosphorylation and on the posterior cell cortex. Conservation of both the kinase and the carboxy-terminal 100 amino acids in proteins from yeast to man hints at possible conservation of function.

This paper reports the distribution of the PAR-3 protein in early embryos, and presents evidence that PAR-3 may function to coordinate cytoplasmic polarity with orientation of the mitotic apparatus. Results include the finding that PAR-3 protein is localized asymmetrically at the cortex in a pattern that is the reciprocal of PAR-1, that PAR-1 depends upon PAR-3 to be restricted to the posterior, and that the distribution of cortical PAR-3 within a cell correlates with the cell's ability to reorient its mitotic apparatus.
