26. To isolate nucleosomes positioned at the Nco I end, reconstitutes were incubated in the presence of 10 U of Hae II restriction enzyme in SP6 transcription buffer at 20°C for 45 min and separated from free DNA and differently positioned cores in preparative nucleoprotein gels.
