The first steps in the biosynthesis of the very complex mycobacterial cell wall are described in this paper. The work described involves the synthesis of the unique linker disaccharide (Figure 1) on a polyprenyl P-P- carrier (Figure 5) followed by the assembly of the galactan unit. Assays are also defined for screening of new anti-tuberculosis drugs active against cell wall synthesis.

The antigen 85 complex was described previously as a major secreted protein of M. tuberculosis, implicated in disease pathogenesis through its fibronectin-binding capacities. This is the first evidence that describes the function of the protein as a mycolyltransferase involved in the final stages of cell wall assembly.

In this novel study, cloning and sequencing strategy of the emb region that confers resistance to Emb when expressed in M. smegmatis is described. Evidence also points towards embA and embB genes that encode for arabinosyl transferase that is the primary target for Emb.
