J Immunol 1996, 157:2795-2803. Human CD1 has been shown to present microbial lipids and lipoglycans (LAM) and mycolic acids to T cells. Polyprenylphosphate-pentoses in mycobacteria are synthesized from 5-phosphoribose pyrophosphate
-
Scherman MS, Bournonville LK, Bush D, Xin Y, Deng L, McNeil M: Polyprenylphosphate-pentoses in mycobacteria are synthesized from 5-phosphoribose pyrophosphate. J Biol Chem 1996, 271:29652-29658. The biosynthetic pathway for the formation of polyprenylphosphate-ribose is reported in this important work. 5-phosphoribose pyrophosphate was is also found to be a precursor of polyprenylphosphate-arabinose (DPA) and the latter is formed via 5P-DPA.

Biosynthesis of the linkage region of the mycobacterial cell wall
-
Mikusov K, Mikus M, Besra GS, Hancock I, Brennan PJ: Biosynthesis of the linkage region of the mycobacterial cell wall. J Biol Chem 1996, 271:7820-7828. The first steps in the biosynthesis of the very complex mycobacterial cell wall are described in this paper. The work described involves the synthesis of the unique linker disaccharide (Figure 1) on a polyprenyl P-P- carrier (Figure 5) followed by the assembly of the galactan unit. Assays are also defined for screening of new anti-tuberculosis drugs active against cell wall synthesis.

Early steps in the biosynthesis of LAM
-
Besra GS, Morehouse CB, Rittner CM, Waechter CJ, Brennan PJ: Early steps in the biosynthesis of LAM. J Biol Chem 1997, 272:18460-18466. In this elegant study, origins of the mannose units of the PIMs and LM of mycobacterial lipoglycans have been described. In addition to this, a linear 16 linked linear mannan was also isolated and identified to be an apparent precursor of mature LM and LAM.

Role of the major antigen of Mycobacterium tuberculosis in cell wall biogenesis
-
Belisle JT, Vissa VD, Sievert T, Takayama K, Brennan PJ, Besra GS: Role of the major antigen of Mycobacterium tuberculosis in cell wall biogenesis. Science 1997, 276:1420-1422. The antigen 85 complex was described previously as a major secreted protein of M. tuberculosis, implicated in disease pathogenesis through its fibronectin-binding capacities. This is the first evidence that describes the function of the protein as a mycolyltransferase involved in the final stages of cell wall assembly.

The embAB genes of Mycobacterium avium encode an arabinosyl transferase involved in cell wall arabinan biosynthesis that is the target for the antimycobacterial drug ethambutol
-
Belanger AE, Besra GS, Ford ME, Mikusov K, Belisle JT, Brennan PJ, Inamine JM: The embAB genes of Mycobacterium avium encode an arabinosyl transferase involved in cell wall arabinan biosynthesis that is the target for the antimycobacterial drug ethambutol. Proc Natl Acad Sci USA 1996, 93:11919-11924. The molecular mechanisms of drug action and drug resistance of the antimycobacterial compound, ethambutol (Emb), are unknown. In this novel study, cloning and sequencing strategy of the emb region that confers resistance to Emb when expressed in M. smegmatis is described. Evidence also points towards embA and embB genes that encode for arabinosyl transferase that is the primary target for Emb.
