This key paper describes the identification of a family of enzymes catalyzing phosphopantetheinyl transfer in a variety of biosynthetic pathways and organisms. More than 20 putative phosphopantetheinyl transferases (PP-Tases) were identified by their similarity to the E. coli holo-acyl carrier protein synthase - they all possess two PPTase consensus sequences. PPTase activity was confirmed for the E. coli proteins EntD (involved in enterobactin biosynthesis) and o195 (unknown open reading frame) as well as for the B. subtilis protein Sfp (involved in surfactin biosynthesis), all of previously unknown function. Given the demonstration of multiple PPTases in E. coli, we suggest that each phosphopantetheine-requiring pathway may have its own PPTase.

In this study it is shown that the Streptomyces polyketide synthase acyl carrier proteins (ACPs) required for granaticin, frenolicin, oxytetracycline and tetracenomycin biosynthesis may be phosphopantetheinylated in vitro using E. coli ACP synthase (ACPS) with varying catalytic efficiencies. When these ACPs are overexpressed in E. coli, typically little holo-ACP is obtained [3,4]. It is also demonstrated that ACPS may be used to add altered phosphopantetheine molecules to ACP substrates, potentially permitting the incorporation of unnatural building blocks into the resultant products.

Alkylation of gramicidin S synthetases 1 and 2 with either the corresponding amino acid substrate or with N-ethylmaleimide was followed by proteolysis and subsequent mass spectrometric analysis of the resultant fragments. This demonstrated the presence of multiple 4′-phosphopantetheine groups (one mole for each mole of active site peptide fragment) which are attached to the serine residue in the consensus phosphopantetheinylation site, and which are covalently thiolated with aminoacyl groups. This clearly distinguishes the multiple thiotemplale mechanism of non-ribosomal peptide synthetases from the type II PKS system, which uses a single holo-ACP to shuttle intermediate peptides.

J Bacteriol 1997, 179:705-713. Two genes, snbA and snbC, involved in the synthesis of pristinamycin I in Streptomyces pristinaespiralis, were identified. The predicted sequence of SnbA shows similarity to EntE, the adenylating enzyme which transfers an aryl group to the aryl carrier protein of EntB in the corresponding enterobactin biosynthetic pathway. SnbC appears to be a two-domain peptide synthetase which may activate L-threonine and L-aminobutyric acid. A mechanism similar to that for the initiation of actinomycin biosynthesis [30] is proposed.

Experiments with purified recombinant proteins support a novel mechanism for the biosynthesis of enterobactin in E. coli, in which a 2,3-dihydroxybenzoate (DHB) group is adenylated by EntE and then transferred as a thioester to the phosphopantetheine cofactor of the aryl carrier protein EntB. It is proposed that this aryl group is then transferred to a serine residue attached to a peptidyl carrier protein moiety on holo-EntF to yield enzyme-bound 2,3-DHB-serine, the building block for enterobactin. This model is in contrast with that proposed for actinomycin [30] and pristinamycin [31••] biosynthesis, which suggests a reaction between released aryl-AMP and the 4′-phosphopantetheine thiol, rather than, in this case, enzyme-catalyzed transfer.
