Microtubule polarity can be determined directly for individual microtubules. Centrosome-nucleated microtubules are used to relate microtubule polarity and the arrowhead moire patterns observed by electron cryomicroscopy of microtubules in vitreous ice. The arrowhead patterns are related to superhelical protofilaments and point to the microtubule plus end for right-handed superhelices and to the minus end for left-handed superhelices. Two methods are given to find the protofilament hand.

The interaction site of the microtubule-stabilizing drug taxol is shown in these maps.
