The role of actin-filament capping in Dictyostelium was analyzed by manipulating the expression levels of CapZ, a barbed end capping protein. The most striking observation was that cells which overexpressed CapZ moved faster than wild-type cells, whereas underexpressing cells moved more slowly than wild-type cells.

The function of capping protein in yeast was explored by examining the effect on actin-filament assembly of deleting CapZ, and by identifying genetic interactions that correlated with actin-assembly defects. In yeast, CapZ appears to be important for stabilizing actin filaments from depolymerization, contrasting with the destabilizing effect of CapZ in Dictyostelium. The different cellular roles of CapZ may reflect the need to stabilize actin filaments in the relatively actin-poor cytoplasm of the yeast cell.

This permeabilized cell system was used to test the role of polyphosphoinositides and GTP-binding proteins in regulating actin assembly. A signal transduction pathway that mediates the cytoskeletal response to thrombin-receptor activation was uncovered.

In a series of papers, starting in 1988, Forscher and coworkers have used differential interference contrast imaging to describe retrograde flow of actin in Aplysia growth cones, and to determine the role of retrograde flow in growth cone motility. In this study, they used pharmacological inhibitors to show for the first time that actin flow is driven by myosin activity, confirming an idea that they first suggested in 1988. The identity of the myosin(s) that drive flow is currently unknown.

These authors used CALI to investigate the role of a class V myosin and a class I myosin in growth cone motility. In this technique, a pulse of light is used to inactivate a protein in vivo, after injecting the cell with antibodies to the protein of interest that have been conjugated with a chromophore. CALI is complementary to genetic approaches in that the immediate effects of protein inactivation can be followed by using both approaches. The authors found that inactivation of myosin-V leads to cessation of filipodium extension, whereas inactivation of myosin-I leads to lamellipodium extension.

The mechanism of cortical patch motility was explored by examining the requirement for myosin motor proteins.

The movement of yeast cortical actin patches was visualized using actin-green fluorescent protein (GFP), Sac6-GFP and Abp1-GFP fusion proteins. Metabolic inhibitors inhibit patch movement, suggesting that the movement is not based on random diffusion.
