This paper disagrees with [18] over the polarity of the staining pattern seen in electron micrographs of tubulin sheets grown from the ends of flagellar axonemes and decorated with kinesin heads in the presence of AMP-PNR The plus and minus ends of axonemes were distinguished by studying examples either with brain tubulin extensions of clearly different lengths assembled from both ends, or with extensions assembled only from the plus end using a mixture of NEM-treated tubulin and untreated tubulin. To understand microtubule-associated motility, it is important to know the correct structural polarity of microtubule-motor complexes.

Cryo-electron microscopy and a back-projection method of image analysis [35] are used to obtain the 3-D structures of 11-, 12-, 14- and 15-protofilament microtubules decorated with ncd motor domains. The method can calculate 3-D maps even for structures whose helical symmetry is broken by lattice discontinuities (seams) and reveals the presence or absence of seams. The result is particularly useful for future work, in that it shows that helically symmetrical brain microtubules can be identified.

The result confirmed that of Mitchison [44]. Both results are useful in helping to identify β-tubulin as the monomer that binds to the broader top part of a motor domain.

In this paper, the position of a peak in a difference map comparing two-dimensional crystals of unmodified zinc-tubulin [13] and subtilisin-digested tubulin, which lacks part of the carboxy-terminal segment, supports the identification in [13] of the predicted carboxy-terminal α helix. The plus-minus orientation of zinc-tubulin protofilaments is found by comparison of the projected density with that of protofilaments in normal tubulin sheets growing from Chlamydomonas axonemes. Building on this, the assumption that the intersubunit connection with the highest density in projection represents the heterodimer bond leads the authors to conclude tentatively that the subunit towards the plus end is α-tubulin.
