J Biol Chem 1996, 271:7336-7342. The authors were interested in exploring the importance of heme binding in regulating the dimerization and catalytic activity of nitric oxide synthases (NOS). Gel filtration chromatography yielded two different elution volumes for heme-saturated and heme-deficient NOS, but these data alone were not sufficient to establish whether the difference was due to conformational changes or dimerization. Static light scattering, in which only the relative intensity of the two solutions at a single scattering angle was measured, clearly showed that the molecular masses of heme-deficient and heme-saturated NOS differed by a factor of two. This shows that significant issues can be resolved with relatively straightforward experiments.

Activation energies were calculated from changes in self-association kinetics with temperature. Characteristics of the equilibrium size distributions following initiation of self-association were measured by static light scattering. A mechanistic description of the self-association pathway was derived based on the data.

However, static light scattering intensity measurements were substantially more sensitive to these changes, and report on peptide binding with surprising accuracy. The data showed that the colicin E1 channel peptide penetrated into the hydrocarbon core of the bilayer in the closed channel state.

Initially, the proteoglycan is modelled as a simply branched polymer undergoing one (or more) of three different degradation mechanisms. Static light scattering measurements taken as a function of time during the degradation process were analyzed to identify the key degradative mechanism for each of the three different degradation methods, and to quantitate degradation rate constants.
