Mutations in the cardiac myosin binding protein-C gene on chromosome 11 cause familial hypertrophic cardiomyopathy
-
Watkins H, Conner D, Thierfelder L, Jarcho J, MacRae C, McKenna W, Maron B, Seidman J, Seidman C: Mutations in the cardiac myosin binding protein-C gene on chromosome 11 cause familial hypertrophic cardiomyopathy. Nat Genet 1995, 11:434-437. Two unrelated families were found to show linkage to the CMH4 locus. A splice donor mutation in the MyBP-C gene was identified in one family, and a duplication mutation was identified in the other. Both mutations were thought to be likely to interfere with myosin heavy-chain binding.

Cardiac myosin binding-C gene splice acceptor site mutation is associated with familial hypertrophic cardiomyopathy
-
Bonne G, Carrier L, Bercovici J, Cruaud C, Richard P, Hainque B, Gautel M, Labeit S, James M, Beckmann J, et al.: Cardiac myosin binding-C gene splice acceptor site mutation is associated with familial hypertrophic cardiomyopathy. Nat Genet 1995, 11:438-440. The authors report a mutation in the MyBP-C gene in two related French families previously linked to the CMH4 locus. They propose that the mutation most likely results in a truncated protein product or a null allele.

Mutations in either the essential or regulatory light chains of myosin are associated with a rare myopathy in human heart and skeletal muscle
-
Poetter K, Jiang H, Hassanzadeh S, Master S, Chang A, Dalakas M, Rayment I, Sellers J, Fananapazir L, Epstein N: Mutations in either the essential or regulatory light chains of myosin are associated with a rare myopathy in human heart and skeletal muscle. Nat Genet 1996, 13:63-69. Missense mutations in the ELC and RLC of myosin are described and are found to be associated with a rare HCM phenotype. In vitro studies of myosin with these mutations are reported. The mutations are suggested to impair the stretch activation response of cardiac papillary muscle.

Altered cardiac troponin T in vitro function in the presence of a mutation implicated in familial hypertrophic cardiomyopathy
-
Lin D, Bobkova A, Homsher E, Tobachman L: Altered cardiac troponin T in vitro function in the presence of a mutation implicated in familial hypertrophic cardiomyopathy. J Clin Invest 1996, 97:2842-2848. Rat cardiac troponin T containing the He79Asn mutation was found to have 50% faster thin filament movement than normal. The relationship between this and altered cardiac contraction in patients with HCM is discussed.
