In human cells, metaphase chromosomes are arranged in a ring or 'rosette'. The authors of this paper used FISH to demonstrate a new level of nuclear architecture where homologous chromosomes are located on opposite sides of the rosette in metaphase. This arrangement is maintained in telophase and may represent a general feature of nuclear organization.

All loci tested occupied different discrete subregions in the nucleus. A subset was found that specifically associated with the nuclear envelope.

By using a green fluorescent protein tagged centromere protein, centromeres were tracked in living HeLa cells. During interphase, most centromeres appeared relatively immobile but some underwent persistent directed motions lasting many minutes and covering over a micron in distance. This result provides the first direct evidence for a motor-like activity within the interpase nucleus.

Demonstration, using FISH, that insertion of a heterochromatic satellite repeat at the Brown locus in Drosophila causes that locus, which is near the end of the chromosome, to loop back and associate with the centric heterochromatin. This result suggests that changes in nuclear position correlate with, and perhaps cause, changes in transcriptional repression of genes undergoing PEV.
