Predicting coiled coils by use of pairwise residue correlations

The application of pairwise residue correlations to the problem of coiled-coil detection results in a method that shows an excellent resolution between long parallel coiled coils and globular proteins, but that loses the ability to pick up coiled coils whose sequences are far removed from the coiled-coil consensus, indicating that the method is overly stringent. of outstanding interest

An ingenious method to include backbone motions into conformational searches by algebraic parametrization. Although the method is only used to reproduce the coordinates of the GCN4 dimeric, trimeric and tetrameric mutants in this paper, it clearly has the potential to simplify and, at the same time, make the search for favourable sidechain conformations in the modelling of coiled-coil cores more accurate. of special interest

A peptide fragment of human DNA topoisomerase II a forms a stable coiled-coil structure in solution
-
In this paper, the authors apply a combination of secondary-structure and coiled-coil prediction to identify a candidate coiled-coil helix in topoisomerase II, then test their prediction against the isolated peptide. Considering the difficulty in interpreting the prediction data, the accuracy with which the authors identify the helix, including the location of its ends, is impressive. of special interest

Hyperthermostable surface layer protein tetrabrachion from the archaebacterium Staphylothermus marinus: Evidence for the presence of a right-handed coiled coil derived from the primary structure
-
The incredible coiled-coil stalk of this surface-layer protein combines some truly unusual properties - a left-handed coiled coil that switches to a right-handed conformation at an internal proline, resistance to temperatures up to 125°C in 1% SDS or 6 M guanidinium hydrochloride; a disulphide ring, binding sites for two globular protease particles, and multiple glycosylation sites - which convert it into a 'candy stick' of sorts. This protein represents the first clear candidate for a right-handed coiled coil. of outstanding interest
